Where does it state formally that once a driver inserts its digi card into the slot first 15 mins of other work should be recorded before any other activity? These 15 mins must stand for daily vehicle checks.

It doesn’t state it anywhere.
It states it over there =========> in most RDC waiting rooms.
There isn’t a legal requirement to show any amount of work before driving, it’s more of an interpretation of rules to provide a defence.
What you have to consider is if a defect was found on your vehicle during a road-side check, if you had put your card/chart in and driven off straight away, how would you convince the DVSA inspector that you had set off on your journey with the vehicle in a roadworthy condition?
That is why many companies now insist that 15 minutes work is recorded prior to setting of, so they can demonstrate that pre-journey checks are being carried out.
Is it 15 mins of other work done by manual entry or it has to be recorded once the card is inserted?

The 15 minutes is just a company thing so it would be what they want you to do.
I would always do it as a manual entry, no point in inserting your card, checking the vehicle then finding something wrong that means the vehicle is VIR and you have to take a different truck. Once I know it’s okay to drive I would put the card in and make a mental entry for that period.

It’s scratched into the wall in every RDC waiting room

That’s exactly what I do. I’ve had regs quoted at me in the past stating you must insert the card as soon as you take control of the vehicle blah blah but if you do a manual entry to cover the period spent checking the vehicle I fail to see the problem as you’re not hiding work, it’s all on the card one way or another.
